Famciclovir has ~77% oral bioavailability (vs ~5% for oral penciclovir directly, and ~15\u201320% for oral aciclovir). Inside herpes-virus-infected cells, penciclovir triphosphate blocks viral DNA polymerase \u2014 the same final mechanism as aciclovir.<\/p>\n<p>The clinical edge famciclovir offers is <strong>intracellular persistence<\/strong>: penciclovir triphosphate has a half-life of 10+ hours inside infected cells, vs ~1 hour for aciclovir triphosphate. This lets famciclovir use less-frequent dosing.
