Vriligy contains dapoxetine, the first and only prescription medication developed specifically for the treatment of premature ejaculation (PE). Unlike erectile-dysfunction drugs, dapoxetine does not treat weak erections — it extends intravaginal ejaculatory latency time (IELT) in men with lifelong or acquired PE.

Manufactured by Centurion Laboratories, Vriligy is a short-acting selective serotonin reuptake inhibitor (SSRI) engineered with a very short half-life — so it can be dosed on-demand before intimacy rather than taken daily as a standard SSRI.

What Is Vriligy?

Vriligy is a prescription short-acting SSRI used on-demand to extend ejaculatory latency in men with PE. It is the active ingredient in Priligy®, the first PE medication approved in Europe.

PE is defined clinically as ejaculation that occurs within about one minute of vaginal penetration and causes distress to the man or his partner. Dapoxetine is indicated for men with:

  • Lifelong PE — present from the start of sexual life
  • Acquired PE — developed after a period of normal function
  • Co-existing ED and PE — where dapoxetine is combined with a PDE5 inhibitor under medical supervision

How Does Vriligy Work?

Ejaculation is a reflex coordinated by serotonergic neurons in the spinal cord and brainstem. Dapoxetine works by:

  • Selectively inhibiting the serotonin transporter (SERT) at central nerve terminals
  • Raising synaptic serotonin levels, which delays the ejaculatory reflex
  • Acting quickly (onset ~1 hour) and clearing quickly (half-life ~1.5 hours for the parent compound) — so the effect is confined to the sexual encounter rather than persisting all day

In clinical trials, dapoxetine typically extends IELT by 2 to 3 times the baseline, with responders reporting improved control, reduced distress, and higher partner satisfaction.

Dapoxetine is not an ED drug. If you have both ED and PE, a PDE5 inhibitor is required alongside dapoxetine — either as separate tablets or as a combination product such as Super Tadalis Sx or Super Vilitra.

Uses and Indications

  • Premature ejaculation (PE) — lifelong or acquired, in men aged 18–64
  • Combined ED and PE — co-administered with a PDE5 inhibitor (specialist oversight recommended)

Vriligy Dosage and Administration

ScenarioRecommended DoseFrequency / Timing
PE — starter dose30 mg1–3 hours before intended activity; max 1 dose / 24 h
PE — inadequate response60 mgStep up after 4 weeks if 30 mg gives suboptimal response; max 1 dose / 24 h
Elderly (65+)Not recommendedSafety data insufficient above age 64
Mild hepatic impairment30 mg, max once dailyNo dose escalation
Moderate–severe hepatic impairmentContraindicatedAvoid entirely
Severe renal impairmentNot recommendedInsufficient data
Maximum daily dose60 mg per 24 hoursOn-demand only — not for daily SSRI use

How to Take Vriligy Properly

  • Swallow the tablet whole with a full glass of water — reduces the risk of dizziness and syncope
  • Take 1–3 hours before anticipated activity
  • Can be taken with or without food
  • Do not take more than one dose in 24 hours
  • Avoid alcohol — the combination sharply increases dizziness, syncope risk, and impairs judgement
  • If you feel dizzy or faint, lie down immediately to prevent injury

Side Effects of Vriligy

Dapoxetine has a distinctive side-effect profile — different from PDE5 inhibitors.

SeveritySide Effect
Common (≥1 in 10)Nausea, dizziness, headache
Common (≥1 in 100)Dry mouth, fatigue, diarrhoea, insomnia, anxiety, somnolence
UncommonSyncope (fainting), orthostatic hypotension, sinus bradycardia
RareSerotonin syndrome (mainly with MAOIs or other serotonergic drugs), mania, severe allergic reactions

Syncope warning: 0.05–0.1% of patients experience transient syncope within the first hour. Always take dapoxetine with a full glass of water, sit or lie down if you feel lightheaded, and avoid situations where sudden dizziness could cause harm.

Warnings and Precautions

  • Do not combine with MAOIs (phenelzine, tranylcypromine, linezolid, moclobemide) — risk of serotonin syndrome. Allow 14 days washout after stopping an MAOI
  • Do not combine with other serotonergic agents — SSRIs, SNRIs, tricyclics, tramadol, St John’s wort, triptans — without specialist advice
  • Cardiac conditions — avoid in significant heart disease (heart failure NYHA II+, heart-block, coronary disease, arrhythmia)
  • Mental-health history — avoid in mania, bipolar disorder, or severe depression
  • Dehydration / volume depletion — worsens syncope risk; hydrate before dosing
  • Driving — dapoxetine can cause dizziness and drowsiness. Do not drive or operate machinery for 24 hours after dosing

Contraindications — Who Should NOT Take Vriligy

  • MAOI use within the last 14 days
  • Thioridazine use
  • History of mania or severe depression
  • Significant structural cardiac disease (heart failure, ischaemia, valvular disease, arrhythmia)
  • Moderate–severe hepatic impairment
  • Age <18 or >64
  • Hypersensitivity to dapoxetine

What to Do in Case of Overdose

Dapoxetine overdose can cause severe nausea, dizziness, hypotension, syncope, and in rare cases serotonin syndrome (agitation, tremor, tachycardia, hyperthermia). Management is supportive. Contact your local poisons service or emergency department immediately.

Frequently Asked Questions

Is Vriligy an erectile dysfunction drug?

No. Dapoxetine treats premature ejaculation, not erectile dysfunction. If you have both ED and PE, you need a PDE5 inhibitor in addition to dapoxetine — either separately or as a combination tablet.

How quickly does Vriligy work?

Peak plasma is reached in about 1 hour. Take 1–3 hours before intended activity for best results.

How long does Vriligy last?

The active window is roughly 3–5 hours. The parent compound has a half-life of ~1.5 hours, but an active metabolite extends the effective duration.

How much longer will I last on Vriligy?

Clinical trials show that Vriligy typically extends intravaginal ejaculatory latency time (IELT) by a factor of 2 to 3. Real-world results vary with baseline severity, partner dynamics, and psychological factors.

Can I take Vriligy every day?

No — it is an on-demand medication only. It is not suitable as a continuous daily SSRI.

Can I combine Vriligy with a PDE5 inhibitor?

Yes, under medical supervision. Additive blood-pressure effects mean this combination should be trialled cautiously. Combination tablets such as Super Tadalis Sx and Super Vilitra exist specifically for this use case.

Can I drink alcohol with Vriligy?

No — avoid. Alcohol sharply increases syncope, dizziness, and impaired judgement.

Will Vriligy help psychological or “performance anxiety” PE?

Dapoxetine addresses the physiological side of ejaculatory reflex control. Anxiety-driven PE often benefits from combined pharmacological and behavioural approaches (stop–start technique, pelvic-floor exercises, sex-therapy).

Is Vriligy addictive?

No — dapoxetine is not addictive. It has no abuse potential and does not cause dependence.

What if Vriligy doesn’t work?

Give 30 mg a 4-week trial before stepping up to 60 mg. If 60 mg is still inadequate, options include behavioural therapy, topical anaesthetics (lidocaine/prilocaine cream), or specialist referral.
